A stochastic model of compartmentalized intracellular signalling

Plasma membrane receptors are now recognized to signal both at plasma membrane and within very dynamic intracellular organelles called endosomes. Receptors are moving from one place to another through tightly regulated endocytosis/recycling processes. Those distinct spatial location appear as a key regulatory mechanisms for cells to decode exracellular signals and transduce them as physiological responses. The paradigm of ”bulk” mass-action models of signaling pathways [1] thus need to be modified to represent highly compartmentalised systems. We have build a dynamical model that incorporate endocytosis/recycling events within a signaling model, able to represent both receptor trafficking in endosomes and reaction networks embedded in signaling pathways. This new methodology is formulated as either partial differential equations (PDE) [2] or piecewise deterministic Markov processes (PDMP), which calls for interesting new developments. In particular, as a first step towards a better understanding of those models and their application, we have studied the long time behavior of the model and shown the existence and uniqueness of a stationary measure under mild conditions. We will illustrate those results through several examples.
