Towards controlling data driven models of tumour microenvironments
Given the recent discoveries about the importance of specific interactions between immune and cancer cells, understanding the spatial properties of tumors at single-cell resolution becomes crucial. We have developed computational tools to describe spatial patterns in tissues using network theory to extract statistical properties from spatial-omics to be used as clinical biomarkers (tysserand, Coullomb & Pancaldi 2021, mosna, Coullomb et al. 2026).
We are now turning into studying dynamics in the tissues by coupling our spatial metrics with models of cellular process and phenotype transitions. We propose a combination of agent-based (Verstraete et al. 2023) and logic modeling (Marku et al. 2020) using the PhysiCell ecosystem to represent intra and inter-cellular processes in tumors. We aim to make the models patient-specific and clinically relevant by proposing data-driven approaches.
We will discuss our current attempts to construct such models, including reinforcement learning based approaches to control these systems at least in-silico.
